Overview
ARA-290 is a synthetic research peptide, an 11-amino acid engineered to mimic the three-dimensional structure of the helix B surface domain of erythropoietin (EPO). ARA-290 peptide is non-erythropoietic, designed to retain neuroprotective and anti-inflammatory activities while eliminating hematopoietic function. This Peptide has received Orphan Drug Designation from FDA for treatment of neuropathic pain in sarcoidosis patients and has been investigated for retinal diseases.
Mechanism of Action

ARA-290 exerts its effects through selective, high-affinity binding to the Innate Repair Receptor (IRR)—a heterodimeric receptor complex composed of the erythropoietin receptor (EPOR) and the β-common receptor (βcR/CD131). This receptor is typically expressed at low levels in healthy tissues but is upregulated in response to injury, inflammation, or metabolic stress. By selectively activating the IRR without interacting with the erythropoietic EPO-R homodimer, ARA-290 triggers a cascade of intracellular signaling events that collectively suppress inflammation, inhibit apoptosis, and promote tissue regeneration. Key downstream pathways include JAK2/STAT, PI3K/Akt, and inhibition of the NF-κB inflammatory pathway.
