Overview
IGF-1 LR3 is a synthetic research peptide, an 83-amino acid polypeptide engineered as a lengthened analog of human IGF-1. IGF-1 LR3 peptide features an arginine substitution at position 3 and a 13-amino acid N-terminal extension, resulting in a markedly prolonged half-life of approximately 20–30 hours. This IGF-1 LR3 analog exhibits potent mitogenic and anabolic properties widely studied in cell growth, differentiation, and metabolic regulation research.
Mechanism of Action

IGF-1 LR3 exerts its effects by binding to the Type 1 IGF receptor (IGF-1R), which is homologous to the insulin receptor. This receptor activation stimulates intracellular signaling cascades, primarily the PI3K/AKT pathway and the MAPK/ERK pathway (including ERK1/2 signaling). These pathways promote cell proliferation, survival, differentiation, and metabolic processes including the uptake of glucose, fatty acids, and amino acids to support growing tissues. Unlike native IGF-1, IGF-1 LR3 has >100-fold reduced affinity for IGF-binding proteins (IGFBPs), which otherwise sequester native IGF-1 and limit its bioavailability. This structural advantage ensures sustained receptor engagement and enhanced biological potency—approximately three times more potent than native IGF-1.
