PNC 27

PNC-27 is a synthetic 32-residue chimeric anticancer peptide composed of an HDM-2 binding domain corresponding to residues 12–26 of the p53 protein and a cell-penetrating peptide (CPP) leader sequence. The compound belongs to a class of peptides designed to selectively target cancer cells while sparing normal, untransformed cells. PNC-27 has demonstrated potent antitumor activity against a wide range of cancer cell lines, including acute myeloid leukemia, cervical squamous cancer, ovarian cancer, colon cancer, and pancreatic carcinoma.

PNC-27 exerts its selective anticancer effects through a unique dual-targeting mechanism. The peptide binds to the p53 binding site of HDM-2 (residues 1–109), a protein that is uniquely expressed on the plasma membranes of cancer cells but not on normal cells. Following binding, PNC-27 induces the formation of transmembrane pores, resulting in the extrusion of intracellular contents and cancer cell necrosis. Additionally, PNC-27 enters cancer cells and binds to mitochondrial membranes, causing mitochondrial disruption. The peptide exhibits selective cytotoxicity across a broad range of cancer types while demonstrating no effect on normal cells.
